01 / Overview

What is Plant Tissue Culture?

Genomic DNA is extracted from leaf, callus or seed tissue and checked for quantity and purity. A chosen DNA sequence — a transgene, a marker gene or a GM element — is then amplified by PCR, and the products are separated on an agarose gel, where a band of the expected size shows the target is present.

Because PCR detects the sequence itself, transformation or GM status can be confirmed from a small piece of tissue, long before plants mature. Consultation support covers media, sterilisation, explant choice and regeneration for tissue culture work.

    Tissue is sampled

    Leaf, callus, seed or plantlet tissue is collected and sent fresh, frozen or dried.

    DNA is extracted

    Cells are broken open and genomic DNA is purified, then checked for quantity and purity.

  3. 03

    Target is amplified

    Primers for the gene, marker or GM element are copied over repeated heating and cooling cycles.

    Bands are read

    Products run on an agarose gel; a band of the expected size against a ladder confirms the target.

Around 100–500 mg tissue per sample; a few grams for seed or grain

Chilled or frozen, sealed and individually bagged, within 48 h
Primers & controls
Primer sequences, expected band size and a positive control if available
